These data thus led us to propose a model in which YAP function in Müller cells would be necessary for ensuring the maintenance of Müller cell homeostasis and preserving cone integrity (Fig. 8). Although we know that Müller glia can function as retinal stem cells following injury, no one has tested whether damage specifically targeted to retinal ganglion cells, as occurs in glaucoma, is sufficient to stimulate Müller glia-dependent retinal ganglion cell regeneration. Müller cells are the most abundant type of glial cells found in the vertebrate retina, and perform diverse functions that support the activity of retinal neurons. This immortalized cell line facilitates gene expression studies in Müller cells and improves the understanding of Müller cell-neuron interactions. Muller cells provide homeostatic and metabolic support for retinal neurons, like the ganglions and starburst amacrines we map in Eyewire. They are retinal glial cells that wrap around the cells that form capillaries, endothelial cells and pericytes, and constitute the majority of the non-neuronal volume of the retina. These cells, Müller cells, are radial glial cells in the inner vertebrate retina, which have a cylindrical, fiber-like shape (their original name was “radial fibers of Müller”) (10). They fulfill a wide range of physiological functions to support the functioning and survival of retinal neurons (11).

Unactivated microglia, labeled vitally with Calcein-AM ( left panel , in green), were seeded onto Müller cells following 48-hr co-culture and allowed to adhere ( right panel , Müller cells seen in bright-field). These data are consistent with our findings, because they argue against the small and transient levels of Müller cell death being the cause of the reduced synaptic function and structural defects observed, and suggest instead that harmonin in Müller cells has a paracrine effect on cone synapse stability, such that the sustained absence of this protein results in the observed dysfunction and progressive photoreceptor cell death. Purpose. Muller cells are thought 10 have an important role in regulating retinal microvascular function and growth. Our goal is to identify the factors produced and released by Muller cells to mediate their function on target cells.

Radial glial cells, or radial glial progenitor cells (RGPs), are bipolar-shaped progenitor cells that are responsible for producing all of the neurons in the cerebral cortex.RGPs also produce certain lineages of glia, including astrocytes and oligodendrocytes. Their cell bodies reside in the embryonic ventricular zone, which lies next to the developing ventricular system. 2011-11-01 2011-12-07 cells Article Electrical Stimulation Induces Retinal Müller Cell Proliferation and Their Progenitor Cell Potential Sam Enayati 1,2,3,4, Karen Chang 1, Hamida Achour 1,4, Kin-Sang Cho 1, Fuyi Xu 5, Shuai Guo 1, Katarina Z. Müller cells express numerous voltage-gated channels and neurotransmitter receptors, which recognize a variety of neuronal signals and trigger cell depolarization and intracellular Ca2+ waves. Müller cells, the major type of glial cells in the retina, are responsible for the homeostatic and metabolic support of retinal neurons.
